293 Serum-Free Medium: Breakthrough Technology Facilitating Biomedical Research and Production

Time:2025-09-23View:

In recent years, with the rapid development of gene therapy, recombinant protein drugs and viral vector vaccines, the HEK 293 cell line has become one of the most important tool cells in the field of biopharmaceuticals.

The breakthrough in serum-free culture medium technology is bringing unprecedented reliability, efficiency and large-scale production capabilities to this field.

1. The Importance of HEK293 Cells

The HEK293 cell line is derived from human embryonic kidney cells and is obtained by culturing them with modified type 5 adenovirus DNA fragments. These cells have the characteristics of high transfection efficiency, rapid growth rate, and ease of cultivation.

It not only enables large-scale suspension culture to meet the requirements of large-scale production, but also possesses all the post-translational modification functions of humans, and can produce proteins that are most similar to those naturally synthesized by humans.

Currently, HEK293 cells have become a powerful platform for the production of proteins, vaccines, anti-cancer drugs and recombinant viral vectors. Since 2015, seven HEK-derived products have been approved by the FDA.

2.  Advantages of Serum-Free Culture Media

Traditional serum-containing culture media have problems such as unclear composition, significant batch-to-batch variations, and potential contamination risks. Serum-free culture media have perfectly addressed these issues by precisely blending various nutrients, growth factors, and additives.

Serum-free media have several significant advantages, including precise control of components, elimination of contamination risks, and optimization of cell growth. The characteristic of being chemically component-limited eliminates the interference from unknown components and exogenous factors in serum, making the experimental results more consistent and reliable.

Meanwhile, the serum-free medium reduces the introduction of foreign proteins, facilitating the separation and purification of downstream products and enhancing safety.

3. Application Areas and Performance Data

The serum-free medium has shown excellent performance in various applications:

In the field of virus packaging, the Icosa OptiVitro® 293 serum-free virus packaging medium HE03 is specifically designed for 293 cell virus packaging. It supports the efficient expansion of suspended HEK293T, 293F, etc. and plasmid transfection, and can significantly increase the virus titer.

Studies have shown that by using the optimized serum-free culture medium system, the production of lentiviruses can reach 10⁷ TU/mL, and the production of adeno-associated viruses (AAV) can even reach a high level of 10¹¹ vg/mL.

In terms of protein expression, serum-free media also perform exceptionally well. Certain specialized media support efficient protein expression, and the instantaneous protein expression level in the flow processing can reach 900mg/L.

4. Cell Cultivation and Conditioning Recommendations

Transitioning cells from serum-containing media to serum-free environments requires a scientific cultivation method. A progressive serum reduction strategy is typically employed, reducing the serum concentration from 10% to 0% in 4-6 steps. Each adjustment requires 3-5 generations of culture cycles.

The experts recommend starting the acclimation process with a 50% original medium + 50% serum-free medium mixture ratio, and then gradually increasing the proportion of the serum-free medium. When the cell viability is above 90%, it can be transferred to 100% serum-free medium for cultivation.

The optimal inoculation density is recommended to be controlled within the range of 0.5 - 0.7 × 10^6 cells/mL, and the shaker speed should be maintained at 120 - 140 rpm. This will result in the best cultivation effect.

5. Industry Development and Future Outlook

With the rapid development of the gene therapy industry, the demand for serum-free media is increasing sharply. Since the beginning of 2025, there have been numerous positive developments in the gene therapy field, with multiple IND applications for gene therapy drugs being approved one after another.

The FDA predicts that from 2025 onwards, it will approve 10 to 20 cell gene therapies each year. This will further drive the development of the serum-free culture medium market.

The advancement of serum-free culture medium technology is driving the entire biopharmaceutical industry forward. With the advent of the era of gene therapy, serum-free media, as a key raw material, will become increasingly important. The rise of domestic serum-free media has provided a stable and reliable supply chain guarantee for Chinese biopharmaceutical enterprises. In the future, we anticipate that the serum-free culture medium technology will make further breakthroughs in enhancing cell culture efficiency and reducing production costs, thereby providing more innovative impetus for the biopharmaceutical industry.
